Can I use the multimeter to measure the resistance between Emiter and Colector in the transistors, it should give me 1 since the transistor should be Off, right ?

On this type of transistor, and using your multimeter on the diode scale, you should get ~0.6V between the base (positive of multimeter) and Emitter (Negative of meter). When you reverse the leads, it will read open.
