Hi,
I'm about to add pins for programming an 18LF2520 circuit PCB. The circuit is 3.3V and controlled by a regulator. It has an LCD screen.

For programming if I power the circuit ON, and set to Pickit3 to 3.3V does the +V need to be connected, or will it program with only the 0V connected, plus the programming lines?
Camerart.

If you don't connect Vdd then it reads the voltage and gives an error. So, yes, you need to connect it.
