R2 is a variable resistor(trim pot) , it has 3 legs , the arrow shown is the middle terminal of the variable resistor.

You can use any general purpose, small-signal NPN. I would use 2N3904, but there are many other suitable transistors available.

it just measures/ shows the variation in the skin resistance(aka galvanic skin resistance) .when we lie , biological changes occur in our causes perspiration , so that the skin resistance goes low .

Yes. It needs to be insulated except where connections are made.
